A Study on Microbiological Monitoring in Laboratory Animal

Abstract
Three different microbiological monitoring studies were conducted to examine infection of pathogenic bacteria in experimental animals which were reared in conventional systems or SPF systems. For doing microbiological monitoring. pathogenic
bacteria
including M. pulmonis, P. multocida and P. pnemotropica were isolated and antibodies of six pathogens including M. pulmonis, Mouse hepatitis virus, Sendai vhus, Tizzer's disease, Bordetella bronchiseptica, and Corynebacterium kuscheri were
examined. The
first trial with rats, mice, and rabbits purchased from several sellers was undertaken. The isolation rates of Mypalsmas cultured by PPLO media and identified by Dien's stain were 14% in mice and 62.5% in rats. The infection rats by CF test were
55% in
mice and 75% in rats. The highest CF antibody titers in mice and rats were 1: 10 and 1:8, respectively. The CF antibody was increased in proportion to age. The isolation rates of Pasteurella species according to morphological and biochemical
properities
were 20% in mice and 10% in rats and 5% in rabbits. Among the isolated Pasteurella species, six and two strains of P. pneumotropica were observed in mice and rats. respectively and two stains of P. multocida were indentified in mice, rats and
rabbits
The second trial with mice and rats collected from 7 diferent localities(colonies) was performed. From those animals, positive rates of pathogenic bacteria by serological test with ELISA were 62.4% in Sendai virus, 50.3% in Mouse hepatitis virus,
59.3%
in M. pulmonis and 16.9% in Tyzzer's disease in 7 mouse colonies. Also, positive rates of pathogenic bacteria by serological test with agglutination resulted in 3.2% of Bordetella bronchiseptica and 0% of Corynebacterium kutschceri in 7 mouse
colonies.
The third trial was designed to known the effect of changing enviroment from SPF barried system to conventional system on the SPF animals in terms of infection of microorganism. The changing environment for 6 week from SPF to convention gave rise
to
inducement of several non pathogens including Flavobacterium multivorum, Pseudomonas fluorescence, vesicularis, Staph. Cohnii and Corynebaterium xerosis in the small intestsine and the cecum. However antibodies of Sendai virus, Mouse hepatitis
viurs.
Tizzer's disease, M. pulmonis, Bordetella bronchiseptica and Corynebacterium kutscheri in rats were not produced by shifting environment.
